Should work, see https://musescore.org/en/handbook/tuplets#custom-tuplets. Start the measure with an 8th Rest, range select the remaining rests, Ctrl+5, then change the tuplet's properties to show relation.
Untested...
Not easy since tuplets work best on a whole (ie intact, single entity) note or rest. One way to do it is to jump forward to an empty measure and change the Time Signature to 5/8. Click on the whole rest, [Ctrl]6 to create the sextuplet and then copy and paste and use Inspector to change the tuplet Number type to "Relation".
So, not impossible, but a it would be desirable if more than one rest could be selected and the user could choose whether to a]apply the "tupletization" to each rest individually (a common scenario being changing four quarter-notes to four sets of triplets) or b]apply the tuplet effect to the combined value of the notes or rests selected.

underquark, I don't understand how this works: You create a sextuplet from a half rest inside a 5/8 measure, then copy and paste it into a 3/4 measure. Don't you then get a quarter rest at the start--or an extra eighth rest at the end of the bar? Or how do you get the tuplet to fill the space of 5 rather than 4 quavers?
And even if you can make that work graphically, how do you get playoff to play what is a 5/8 measure in the same time as the (3/4) measures before and after? (if you don't need playoff the trick above will do just fine).

The challenge here was to create 6 eighth notes in the space of 5 and then express the resultant tuplet in terms of the ratio 6:5 but difficulties are encountered because of the way that MuseScore creates tuplets. It creates most tuplets just fine but 6 in in the space of 5 is a less common requirement.
Currently if you select a half-note rest plus an eighth-note rest and try to make a set of tuplets you will find that each rest is treated individually and you get two sets of tuplets. If you want to use the whole, combined, duration then you need a single rest that is of that duration (5 eighths in this case) but you can't create a single rest of 5 eighths duration in a 3/4 measure.
You create a 5/8 measure merely to get the whole-measure rest that is 5 eighth notes in duration. You turn this single rest into 6 thus making 6 notes in the space of 5. You copy from your "donor" 5/8 measure into your recipient 3/4 measure having first placed an eighth beat rest in that 3/4 measure (so you paste onto the second rest). You can then get rid of the 5/8 measure or preserve it somewhere at the end of the piece if you need to use it later.
A useful feature would, therefore, be the ability to select multiple rests and specify whether you want to apply the same tuplet rules to each rest (the commonest scenario) or whther you would prefer to aggregate the rests into one value and apply the tuplet rules only to that value (a less common request but still used in many pieces).
