Increasing this value also increases inter-staff and inter-system distances, so is it a measure of the depth of the skyline above/below each staff?

This setting controls how much space between the skylines. If there was already plenty of room between the staves or systems, increasing this will have no effect. But if there is a spot where a note or marking below one staff or system collides with a note or marking above the next, this settings controls how the amount by which MuseScore will avoid the collision.
