How to create a custom template?
With this can I have saved fonts? Or is there a better way to have default fonts applied to all scores?

Yes. Just set up a score to you likings and then save it in the templates folder

Just like normally save scores, only now in the templates folder

I just saved a file in the defined myTemplates folder and it didn't show up as template - even saving it to C:/Program Files/MuseScore 4/templates (where the default templates are) did not help.
There seems to be more to it than it used to be in the previous version - or I missed something...

I believe you need to restart MuseScore for it to see newly added templates

Nope.
It seems only a newly created score can be used as template in MU4, opening from an older version and converting it does not do the trick.....

This isn't true in general, I have quite a few MuseScore 3 files as templates working fine in MuseScore 4.

Definitely don't save to an internal program folders. It is important to save only to your own folders.
It may also be necessary to restart MsueScore as noted.

If I were trying to create a concert band template, since the one I usually use seems to have been removed, would I just put the template in C:\Program Files\MuseScore 4\templates ? or would I have to go into \07-Band_and_Percussion and put it there?

Weird, I can confirm the standard concert band template seems to be missing, even though it is still present within the source itself. You can apply that fix to your local C:\Program Files\MuseScore 4\templates\07-Band_and_Percussion\01-Concert_Band\01-Concert_Band.mscx
Apart from the template topic:
I would highly recommend using a custom style (define everything under Format, save style in your styles folder) and setting that as default for all scores and parts (Edit - Preferences - Score)
