The portamento function of melodic glissandos does not play when using Muse Sounds. Only beginning and end notes will sound. Using Muse Basic or MS3 resolves the issue.

Hi, when I reported the issue I got linked back to this forum. However, when I try to do the same thing using the Musebasic sound font, I still receive no playback of it.

Happens to me as well
I also had this error—however, while switching to the MS Basic soundfont usually resolved it, something very strange happened with the Tenors voice/instrument: there was no pitch slide, and the first pitch never stopped playing, ultimately sounding like a harmony between the first and second note and then continuing for the rest of playback. Attached is a score which demonstrates all three scenarios.

That sounds like an unrelated issue, best to report it separately. Or add your example to https://musescore.org/en/node/342042.
But do also see https://github.com/musescore/MuseScore/issues/12838 and https://github.com/musescore/MuseScore/issues/15268. The original issue here is already reported on GitHub via those two issues, so I'm closing this as a duplicate.

I have noticed that portamento playback doesn't work when starting from a tied note:

If you need portamento for an instrument where it wasn’t sampled, use a soundfont (or possibly a VST) instead of Muse Sounds, where it can be simulated using digitally.
