Slides on Tablature Staffs
I know you can indicate slides with a line or an 's' on the tab, but how can a slide be recognised in the playback? Thanks

I have used glissando as shown here, however it makes no difference to the playback? Thanks

Glissando playback currently can't do portamento yet: #127341: Add Glissando playback style portamento
As you're notes are only a half tone apart, you indeed won't notice the effect.
Either don't notate the in-between notes and have a gliss between start and end notes or add (invisible) bends for playback purposes.

This doesn't work for sliding down frets (towards the neck). Is there a way to represent this?

As glissandos
Slides along the neck are glissandos (not arpeggios, sorry) after all, on either direction.
Or, maybe, a series of slurred notes.
Bends are sideways and always only increase the pitch
