the play back for a tuba and also trombone have disappeared. the mute buttons have not been engaged.
I have no idea what could have caused this.

It only applies to the a particular piece. Other works have not been affected.

Apparently you figured it out? I see a score attached. But I don't have any trouble hearing the trombone or tuba. In fact, that's all I hear, because the "Solo" option is clicked for those instruments in the Mixer.

As mentioned above, I first reset the notes and dynamics to the default values.

I see you figured out how to attach a score. In the score you uploaded you have the trombone and tuba soloed so they are the only instruments you hear. Press F10 to display the mixer and remove the checks from solo.

@ C.y. Butcher, try it:
Right click on a note, select all similar Elements;
Open Inspector and reset the 'Velocity' (both buttons).
If it does not work attach the score as Jojo said

Certain customizations you may have made in older versions are not compatible with the current version. Just do Help / Revert to Factory Settings, or if you prefer to only reset this in particular, View / Synthesizer, Add, MuseScore_General.sf3, Set as Default.
Could also be that while you didn't mute those instruments, you did solo something else. As mentioned, in order to do more than guess, though, we'd need you to attach your score.
