Is it possible to change key in the middel of a measure (because I have an upbeat)
I have this piece in 4/4 that starts in an upbeat (3/4). So somewhere in the piece the key changes, but is not true for the first beat of the measure, only for the latter 3. Is that possible in Musescore?

No. Just split the measure, then apply the key sig change.
You could even hide the barline, although that might confuse musicians

To expand on the above suggestion, see the attached image.
To make that, I right-clicked on measures 2 & 3, chose "Measure Properties" … under Measure Duration I set "Actual" to 2/4 for each measure, dragged a new key sig into measure #3, and then made the barline invisible with the Inspector. You can still see it but it on-screen as gray but it will not print or be included in a PDF export.
Key changed always happen at the beginning of the measure. Use accidentals for the upbeat.
