Inserting N.C.
Hi,
I have Times Signature of 4/4 but only use quarter note for the first measure. How to remove the rest in the front? Or how to insert the N.C.?

Two ways:
1. The easy way: Use the score creation wizard. On the page where you set the time signature there is choice for an upbeat. Select it and specify a quarter note as the length of he upbeat. Done.
2. If you omitted it at that stage you can right click that first measure and select measure properties. There you change it to 1/4. Check also the box to exclude this partial measure from the measure count, otherwise all measure numbers will be incorrect.

I tried the second option. It works fine but there is additional line between measure.
Can the lines in the red circle be removed?

I think it means "No Chord". But that's just a guess.

I have never seen this additional line. I am afraid I can't help with that. It does not happen to me (I have 2.0.3 on MacOS Yosemite).
What is the measure line too close to? Anyhow you can select the upbeat note and in the inspector you can add leading space or trailing space depending on where you think the score is too crowded.

A pretty clear sign of a corrupt score. Please share the score, and tell us which version of MuseScore on what OS you are using and what the original source of that score is, share that too
The N.C. is just a chord symbol and gets entered just the aame

Except fot the made invisible barlines and the fact that you didn't uncheck the "Don't count measure" button for the anacrusis measure's properties, there's nothing wrong with that file
